Broadly, I am interested in the host-pathogen interrelationship and pathogenesis of infectious diseases. My interests therefore scale from host-selection of pathogens to host-response of infection. Understanding the factors which determine the selection of a suitable host for a particular pathogen and corresponding target cells or biosystems by pathogens has important impacts in controlling widespread pathogens and infections, such as human parvoviruses, human immunodeficiency virus, hepatitis viruses, malaria, tuberculosis and meningitis. It also aids in understanding emerging and re-emerging pathogens like human bocaviruses (HBoV) and influenza. Understanding the pathogenesis of these infections has impacts in drug development and therapeutics, drug resistance monitoring and immunology. Most of the advances in controlling transmission of infectious diseases have come from detailed studies of host-pathogen interactions and epidemiology, yet the connections between these and pathogenesis remain under explored. One of the benefits of working with a multidisciplinary focus is that the questions I ask are amenable to diverse situations, and gives a comprehensive picture disease outcome and predict reliable intervention areas.
The success of such multidisciplinary research depends largely on the quality of tools used in investigations. The development therefore of a variety of strategic research tools like molecular diagnostic methods, genetic and bioinformatic analysis techniques as well as mathematical and computerized models is additional interest.
Current Research
Human parvoviruses: Host-specific factors and mechanisms used by human parvoviruses to interact with, and overwhelm, their mammalian host cells. Parvoviruses are among the smallest known viruses. Until 2005, human parvovirus B19 (B19V) was the only member of this family that infected humans and was associated mostly with hematologic disorders. Discovery of human bocaviruses (HBoV) in 2005 and their association with an entirely different type of infection (respiratory) in humans has called for the need to understand their biology and pathogenesis of these viruses in humans. Various angles of research are being used to elucidate host-selection factors as well as mechanisms used to overwhelm mammalian cells and disease outcomes. Though associated with many disease conditions, human parvovirus specific pathology has been under explored. To this end, my supervisors and I are currently investigating the occurrence of B19V with a very important pathogen causing significant morbidity and mortality in Ghana, Plasmodium falciparum. Though 11.5% prevalence of B19V have been reported from a population of Ghanaian children hospitalized with severe anemia, B19V’s contribution to the condition is unknown. To understand and predict the outcome of anemia from mono and co-infected patients, we are determining pressure imposed by these pathogens on infected patients. We are also determining if there is a relationship between the pattern of seasonality and abundances of the two pathogens. As these two pathogens have individual capacities to cause infections leading to high morbidity and mortality especially in children, their interaction in endemic regions has important public health consequences. We will use data to form the basis to design appropriate prospective and retrospective studies that will aid in understanding spectrum of diseases caused by human parvoviruses (B19V and HBoV). An understanding of the epidemiology and burden will justify further studies in the area.
I
Future Research
Molecular epidemiology and evolution: Advances in scientific tools and pathogen education have resulted in the emergence and re-emergence of new pathogens and drug resistance. The watch on these activities and the rate at which they occur demands immediate and effective measures to contain the spread and impact of diseases. There is a high turn-over of bioinformation. I will perform analysis of some of these databases in an attempt to express emerging and re-emerging patterns of pathogens and their diseases. Additionally, I will build local databases for analyzing location-specific pathogens and their diseases.
Drug discovery: Natural products and their derivatives have traditionally been the most common source of many drugs. Africa is naturally endowed with a lot of these natural products from plants to marine/aqua products. Together with a multidisciplinary research group, I will investigate natural products as targets for drug development. It is anticipated that scientific documentation and prove of the activity of natural products yet explored for use as drugs for infectious diseases will be obtained.

Dr. Pauline Byakika-Kibwika is an infectious diseases physician with special interest in HIV clinical pharmacology. She received her medical degree from Makerere University, Kampala, Uganda. She received a Masters degree in Clinical Epidemiology and Biostatistics and a Masters degree in Medicine (Internal Medicine) from the same university. She is a fourth year registered PhD student of Trinity college Dublin, Ireland under supervision of Dr Concepta Merry.
Dr Byakika and her team have set up a clinical pharmacokinetic laboratory at the Infectious Diseases Institute. She received training in HIV pharmacology and pharmacokinetics at the Department of Pharmacology and Therapeutics at the University of Liverpool and at the Tufts centre for drug development, Boston USA.
She is working with a pharmacokinetic team that aims to answer questions of great public health importance regarding the interaction between antiretroviral, antimalarial and antituberculous drugs.
